تأثیر هشت هفته تمرین استقامتی بر میزان بیان GPX4 و MDA در عضله نعلی رت‌های نر سالمند نژاد ویستار

چکیده
افزایش آهن در اثر افزایش سن در روند فروپتوزیس تأثیر دارد و محصول نهایی پراکسیداسیون لیپیدی، MDA است و GPX4 تنظیم‌کننده مرکزی و البته منفی فروپتوز است. فعالیت‌های ورزشی می‌تواند با کاهش پراکسیداسیون لیپیدی این مسیر سالمندی را تعدیل کند؛ بنابراین هدف از انجام پژوهش، تعیین تأثیر هشت هفته تمرین استقامتی بر میزان بیان GPX4 و MDA در عضله نعلی موش‌های صحرایی نر سالمند بود. این مطالعه از نوع تجربی است. 20 سر موش صحرایی نرسالمند (با میانگین سنی 20 ماهه و میانگین وزنی 373 گرم) به دو گروه 10 تایی تقسیم شد: گروه تمرین، گروه کنترل. تمرین به مدت هشت هفته، پنج جلسه در هفته با تردمیل ویژه جوندگان به‌طور فزاینده انجام شد. 48 ساعت بعد از آخرین جلسه تمرین، عضله نعلی موش‌ها طی جراحی برداشته شد و میزان GPX4 به روش بیان ژن و میزان MDA به روش الایزا اندازه‌گیری شد. برای آنالیز داده‌ها، برای سنجش توزیع طبیعی داده‌ها از آزمون شاپیروویلک و برا تعیین همگنی واریانس‌ها از آزمون لوین و برای تعیین تفاوت بین دو گروه از آزمون t مستقل استفاده‌شده و از آزمون دی کوهن برای اندازه‌گیری اندازه اثر استفاده شد و محاسبات آماری توسط نرم‌افزار Prism Graph Pad نسخه هشت انجام شد. اختلاف معنادار در میزان بیان ژن GPX4 در عضله نعلی موش‌های گروه تمرین نسبت به موش‌های گروه کنترل مشاهده نشد 4079/0(p= ولی میزان MDA در گروه تمرین نسبت به گروه کنترل کاهش معناداری داشت (0028/0p=). بنا بر یافته‌های تحقیـق حاضـر، به نظـر می‌رسد اثر تمرین استقامتی در این پژوهش، کاهش فروپتوز را نشان می‌دهد. عدم اختلاف معنادار میزان بیان ژن GPX4 نشان از عدم تأثیر تمرین استقامتی با این شدت بر آن است.

Introduction: The increase of iron due to aging affects the process of ferroptosis, and the final product of lipid peroxidation, MDA, is the central regulator of ferroptosis. Therefore, the aim of the research was to determine the effect of eight weeks of endurance training on the expression of GPX4 and MDA in the soleus muscle of aged male rats.

Materials and Methods: This study is of an experimental type. 20 healthy rats were divided into two groups of 10 each: training group and control group. The training was done incrementally for eight weeks. 48 hours after the last training session, the soleus muscle of mice was surgically removed and the amount of variables was measured by gene expression and ELISA methods.

Results: There was no significant difference in the level of GPX4 gene expression in the soleus muscle of exercise group rats compared to control group rats (p=0.4079). But the amount of MDA in the training group was significantly reduced compared to the control group (p=0.0028).

Conclusion: According to the findings of the present research, it seems that the effect of endurance training in this research shows a decrease in ferroptosis. The absence of a significant difference in the level of GPX4 gene expression is a sign that endurance training with this intensity does not affect it.
